Im not a big fan of mcaffee. Adaware SE 1.6, Spybot S&D 1.4, spysweeper, spywareblaster, crap cleaner (http://www.ccleaner.com), and yes if its still in Beta MS Antispyware, or Counterspy.

Behind behind a router with NAT for a firewall is good
Run all your windows updates
Install Google Toolbar for Internet Explorer..helps keep those popups to a minimum, plus it's really useful.
Spybot search and destroy, latest is 1.4, free, update frequently, and keep it immunized after each update.
Spyware Blaster is good, free version you have to manually update
Mozilla's Firefox works well for many people as an alternative browser, depending on the websites you frequent, as some don't always work well.
Always change your Administrator password, as many threats utilize a blank password to help install themselves
For antivirus, NOD32 and Kapersky are tops in ad/spy/malware/trojan detection and removal. I'm a fan of NOD32.
I like Microsofts Antispyware.
